Implementation of an improved platform for downstream processing of plasmid DNA

Plasmid DNA (pDNA) represents the backbone of most cell and gene therapy production workflows. As a key raw material, its quality can have an important effect on subsequent downstream processes. The increasing demand for highly purified pDNA at gram scale creates a need for controllable, robust, and scalable processes.

This webinar will provide insights into optimizing a process for capture of pDNA, and showcase data on a novel approach to removal of undesired pDNA isoforms. Attend for the opportunity to hear from purification scientists who will discuss the scientific challenges posed by the development process, resin and membrane screen study data, and their approach to working across different sizes of pDNA contruct while maintaining high yield and purity.

  • How different types of chromatography media perform in capture of pDNA from clarified lysate
  • The impact of size on the performance of chromatographic pDNA capture
  • Use of selective binding of pDNA on hydrophobic chromatography to increase purity and decrease consumption of ammonium sulfate
  • Implementing a platform for purification of any pDNA

Justina Martinkeine
Scientist, Purification Sciences at AstraZeneca

Justina Martinkiene graduated from Vilnius Gediminas Technical University (VGTU) in Lithuania in 2016 with bachelor’s and master’s degrees in bioengineering. During the study period she was working as an engineer in VGTU and was developing a purification methodology for enzymes called inulinases from Helianthus tuberosus. After graduation, she worked as a Junior Analyst at TEVA Sicor Biotech in Lithuania where she was responsible for QC testing of recombinant biopharmaceuticals. In 2019 Justina joined AstraZeneca as an Associated Scientist in the Purification Process Sciences Team, where she is involved in process development activities of various modalities: pDNA, antibodies and mRNA.
